Tagraxofusp

Antineoplastic and Immunomodulating Agents

Tagraxofusp, sold under the brand name Elzonris, is an anti-cancer medication described as a CD123-directed cytotoxin. It is a fusion protein combining interleukin-3 with diphtheria toxin, and it kills cultured plasmacytoid dendritic cells by binding to their interleukin-3 receptors, then blocking protein synthesis through the diphtheria toxin component, which inhibits the eukaryotic elongation factor 2. It treats blastic plasmacytoid dendritic cell neoplasm in patients aged two years and older and is given by intravenous injection. It received United States FDA approval in 2018, as a first-in-class medication with priority review, breakthrough therapy and orphan drug designations, and European Union approval in January 2021.
