Tazemetostat

Tazemetostat is an oral cancer drug that was sold under the brand name Tazverik and worked by selectively inhibiting EZH2, an enzyme that adds methyl groups to histone proteins and thereby controls which genes a cell switches on or off. By blocking both normal and mutated EZH2, it disrupted gene expression patterns that let certain cancers grow. Ipsen's tazemetostat received accelerated approval from the United States Food and Drug Administration in January 2020, with orphan drug status, for adults and adolescents aged sixteen and over with metastatic or locally advanced epithelioid sarcoma that could not be fully removed by surgery, taken by mouth at 800 milligrams twice a day. In its pivotal trial of 62 patients the overall response rate was 15 percent. On March 9, 2026, Ipsen voluntarily withdrew tazemetostat from every market after further trials linked it to secondary blood cancers, including T cell lymphoblastic lymphoma and acute myeloid leukemia, ending its clinical use.
