Vorasidenib

Antineoplastic and Immunomodulating Agents

Vorasidenib is an oral cancer drug sold under the brand name Voranigo that inhibits mutated forms of the enzymes isocitrate dehydrogenase 1 and 2. Tumor cells carrying a mutation in either enzyme produce an abnormal metabolite called 2 hydroxyglutarate that drives malignant growth, and vorasidenib blocks both mutated forms at once, letting affected cells differentiate normally and slowing their growth. Servier Laboratories brought the drug to approval, and the United States Food and Drug Administration cleared it in August 2024 for adults and children twelve and older with grade 2 astrocytoma or oligodendroglioma carrying a susceptible IDH1 or IDH2 mutation who have already had surgery. It was the first FDA approved systemic therapy for that specific patient group. Vorasidenib is taken as a 40 milligram tablet once a day, giving patients with these slow growing brain tumors an oral option after surgery rather than immediate chemotherapy or radiation.
