TNF receptor associated periodic syndrome

Diseases of the Blood and Immune Mechanism

TNF receptor associated periodic syndrome (TRAPS) is an inherited periodic fever syndrome caused by mutations in the gene for a receptor for tumor necrosis factor (TNF), inherited in an autosomal dominant manner. People affected experience recurring episodes of high fever along with rash, abdominal pain, joint and muscle aches, and puffy eyes, with episodes recurring over months or years. The condition arises from mutations in the TNFRSF1A gene, which encodes tumor necrosis factor receptor 1 and is located at chromosomal position 12p13.31; around seventy different mutations of this gene have been linked to the syndrome. Because inheritance is autosomal dominant, a single mutated copy of the gene inherited from either parent is enough to produce the condition. TRAPS is studied and managed within immunology as one of the recognized autoinflammatory periodic fever syndromes.
