Hormonal therapy in oncology is one of the major modalities of cancer pharmacotherapy, alongside cytotoxic chemotherapy and targeted therapy, working by manipulating the endocrine system through hormones or drugs that inhibit hormone production or activity. Because steroid hormones drive gene expression in certain cancer cells, altering hormone levels can slow or stop tumor growth, particularly in hormonally responsive cancers such as breast, prostate, endometrial and adrenocortical cancer, and the approach can also relieve paraneoplastic symptoms such as cancer-related anorexia. Its most familiar example is the selective estrogen-response modulator tamoxifen, used to treat breast cancer, though aromatase inhibitors have taken an expanding role in breast cancer treatment as well.
