Cisplatin Through Its DNA Mechanism, Not Its Cancer List

Cisplatin is usually introduced by the cancers it treats, testicular and ovarian cancer chief among them. That list explains why an oncologist reaches for it, but it says nothing about why the drug works, and the mechanism is a different story worth telling on its own. Cisplatin binds to DNA and inhibits its replication through intrastrand crosslinks with purine bases. A cell copying its DNA cannot read past a crosslink cleanly. The cell's own repair machinery tries to fix the damage, and when repair fails the cell undergoes apoptosis, a death the cell's own internal signal triggers rather than an outside poison. Cisplatin does not attack a tumor the way a scalpel removes one; it exploits the one thing every dividing cell must do, which is copy its DNA, and it does so without distinguishing a cancer cell's DNA from a healthy cell's. That mechanism is what places cisplatin in the platinum-based antineoplastic class, a family defined by this DNA-crosslinking action rather than by which cancer a physician happens to be treating that day. The FDA approval came on December 19, 1978, the date this atlas's own sourced history record for cisplatin gives, marking the point a platinum-coordination compound built on this crosslinking mechanism entered approved cancer care rather than staying a laboratory finding. Cisplatin's most significant risks, bone marrow suppression, hearing problems including severe hearing loss, kidney damage and vomiting, are grouped together for a reason worth reading through the same lens: none of them is a separate toxic side reaction.
