Perindopril is a long-acting angiotensin-converting enzyme inhibitor used to treat high blood pressure, heart failure, and stable coronary artery disease. It is a prodrug, meaning it is inactive as administered and is converted in the liver to its active form, perindoprilat, which then works by inhibiting production of the vasoconstricting hormone angiotensin, relaxing blood vessels, increasing urine output and reducing blood volume, effects that together lower blood pressure and ease the workload on the heart. Perindopril was patented in 1980 and approved for medical use in 1988, and is marketed both on its own, as perindopril erbumine or perindopril arginine salts, and in fixed combinations with other medicines including indapamide and amlodipine. It is a prescription-only medicine in the United States, United Kingdom and Canada, and a triple combination of perindopril, amlodipine and indapamide appears on the World Health Organization's Model List of Essential Medicines. In Australia, perindopril ranked as the fourth most prescribed medication in the 2023 to 2024 period.
