Dynemicin A

Antineoplastic and Immunomodulating Agents

Dynemicin A is an anti-cancer drug of the enediyne class, a natural product of the soil bacterium Micromonospora chersina that was first isolated from soil in the Gujarat State of India. It has a bright purple color from its anthraquinone chromophore, and it was originally isolated for its use as a dye before research turned to its anti-cancer properties. Bristol-Myers elucidated its structure in Japan by X-ray diffraction of a closely related compound, triacetyldynemicin A, and Myers and coworkers reported the first chemical synthesis. It acts on B-DNA by intercalating into the minor groove; once bound, its epoxide is activated and the enediyne undergoes a re-cyclization that cleaves both DNA strands and leads to cell death. The source describes it as promising for cancer treatment but still requiring further research, and its pharmacology is not yet fully explored.
