Fedratinib

Antineoplastic and Immunomodulating Agents

Fedratinib is an oral cancer drug sold under the brand name Inrebic that acts as a semi selective inhibitor of Janus kinase 2, or JAK2, with particularly strong potency against that target. By blocking JAK2 it interrupts the downstream JAK-STAT signaling that drives abnormal blood cell production in myeloproliferative disease, causing affected cells to stop multiplying and die. The United States Food and Drug Administration approved fedratinib on August 16, 2019, granting it both priority review and orphan drug designation, for adults with myelofibrosis, a rare bone marrow cancer that causes scarring of the marrow along with anemia, an enlarged spleen and symptoms such as fatigue, night sweats and weight loss, in patients carrying the genetic mutations associated with the disease. Fedratinib is taken as capsules by mouth, giving myelofibrosis patients a second JAK inhibitor option distinct from the earlier drug ruxolitinib.
