Bcr-Abl tyrosine-kinase inhibitors are the first-line therapy for most patients with chronic myelogenous leukemia, a cancer caused in more than 90 percent of cases by a chromosomal abnormality called the Philadelphia chromosome. That abnormality fuses the Abl tyrosine kinase gene with the Bcr gene to create a constitutively active Bcr-Abl tyrosine kinase, and these drugs were developed to selectively inhibit it.
